874 Remodeling of keratinocyte higher-order chromatin organization during differentiation

Our study aimed to compare insights on the dynamics and functional meaning of remodeling 3D higher-order chromatin structure from studies using microscopy analysis FISH experiments genomic utilizing high-throughput chromosome conformation capture assays. We have performed publicly available data several developmentally controlled loci harboring genes important for keratinocyte differentiation. Big part involved in epidermal differentiation are clustered loci. This clustering is establishment their proper spatio-temporal expression pattern. Previous work showed that mouse one those loci, complex (EDC), almost 70 genes, relocates during development nuclear periphery interior. Another demonstrated same locus human loops out territory cultured cells. re-analyzed Hi-C, Capture-C Hi-ChIP hESCs primary keratinocytes found significant differences contact frequencies at studied loci: Ker I, II EDC. results show reported change earlier undergo rewiring promoter-enhancer interactions associated with big changes gene expression.
